New long-term data reveals PALSONIFY's sustained success in acromegaly treatment

Crinetics Pharmaceuticals has presented new long-term data for its drug PALSONIFYTM (paltusotine) at ENDO 2026. The findings highlight the medication’s effectiveness and tolerability in treating acromegaly. Researchers also explored its use alongside oral cabergoline for improved outcomes. The data came from two open-label extension (OLE) studies, PATHFNDR-1 and PATHFNDR-2. In PATHFNDR-1, patients maintained stable IGF-1 levels at 0.81 times the Upper Limit of Normal (ULN) after 96 weeks of treatment. Symptoms and pituitary tumour volumes also remained unchanged throughout the study.

In PATHFNDR-2, IGF-1 levels dropped from 1.64x ULN at baseline to 0.96x ULN after 72 weeks. Seven of the 83 participants saw a reduction of over 20% in tumour volume within 24 weeks. Median Acromegaly Symptom Diary scores stayed consistent at all assessed points in both trials.

The drug proved well-tolerated for most patients. Only four individuals (2.4%) left an OLE due to adverse effects. The most frequent side effects, occurring in over 10% of the pooled OLE group, included diarrhoea, joint pain, headaches, and urinary tract infections. Additionally, combining PALSONIFY with oral cabergoline further lowered IGF-1 levels in some cases. PALSONIFY demonstrated sustained effectiveness and safety in long-term use for acromegaly. The drug maintained stable hormone levels and tumour sizes in many patients. Its combination with cabergoline may offer additional benefits for certain individuals.
